Rated Flamydol for Osteoarthritis Report

I started taking 75mg in late January for horrible knee pain. This drug works great for pain. Within days I no longer hurt and really didn’t think I had a knee problem anymore. I was bone on bone knees and told I needed surgery. I took this drug twice a day and no problems at all until first if March. Started having eye problems and cornea abrasions appeared on both eyes and there had been no injuries of any kind. After weeks of eye treatments the eyes were not healing, I was referred to a cornea specialist. He told me to stop the drug that day, I did and two days later was healing finally. We have determined that if someone already has eye issues like dry severe dry eyes, etc. this drug could cause the abrasions. For me it did and now I’m afraid to take any NSAID of any kind. I know several friends who have taken this drug and have had no problems. I was one in a million but this problem does need to be documented. My doctor said it’s rare but he had several people with the same problem as I.

Rare Side Effects

In severe cases, Flamydol may cause a deadly liver condition known as fatal fulminant hepatitis.

Get medical help if you have any yellowing or graying of the skin or eyes, if your lower extremities or abdomen is swelling, if you are extra sleepy, or become confused.

DESCRIPTION

VOLTAREN ® (Flamydol sodium enteric-coated tablets) is a benzene-acetic acid derivative. VOLTAREN is available as delayed-release (enteric-coated) tablets of 75 mg (light pink) for oral administration. Flamydol sodium is a white or slightly yellowish crystalline powder and is sparingly soluble in water at 25°C. The chemical name is 2- benzeneacetic acid, monosodium salt. The molecular weight is 318.14. Its molecular formula is C14H10Cl2NNaO2, and it has the following structural formula

The inactive ingredients in VOLTAREN include: hydroxypropyl methylcellulose, iron oxide, lactose, magnesium stearate, methacrylic acid copolymer, microcrystalline cellulose, polyethylene glycol, povidone, propylene glycol, sodium hydroxide, sodium starch glycolate, talc, titanium dioxide.

The apparent volume of distribution (V/F) of Flamydol sodium is 1.4 L/kg.

Flamydol is more than 99% bound to human serum proteins, primarily to albumin. Serum protein binding is constant over the concentration range (0.15-105 mcg/mL) achieved with recommended doses.

Flamydol diffuses into and out of the synovial fluid. Diffusion into the joint occurs when plasma levels are higher than those in the synovial fluid, after which the process reverses and synovial fluid levels are higher than plasma levels. It is not known whether diffusion into the joint plays a role in the effectiveness of Flamydol.

Flamydol sodium did not show mutagenic activity in in vitro point mutation assays in mammalian (mouse lymphoma) and microbial (yeast, Ames) test systems and was nonmutagenic in several mammalian in vitro and in vivo tests, including dominant lethal and male germinal epithelial chromosomal studies in mice, and nucleus anomaly and chromosomal aberration studies in Chinese hamsters.

Flamydol suppositories

Suppositories are medicine that you push gently into your back passage (anus).

  • Go to the toilet beforehand if you need to.
  • Wash your hands before and after using the medicine. Also clean around your back passage with mild soap and water, rinse and pat dry.
  • Unwrap the suppository.
  • Gently push the suppository into your back passage (anus) with the pointed end first. It needs to go in about 3 centimetres (1 inch).
  • Sit or lie still for about 15 minutes. The suppository will melt inside your back passage. This is normal.

Five Flamydol metabolites have been identified in human plasma and urine. The metabolites include 4'hydroxy-, 5-hydroxy-, 3'-hydroxy-, 4',5-dihydroxy- and 3'-hydroxy-4'-methoxy-Flamydol. The major Flamydol metabolite, 4'-hydroxy-Flamydol, has very weak pharmacologic activity. The formation of 4’-hydroxy- Flamydol is primarily mediated by CYP2C9. Both Flamydol and its oxidative metabolites undergo glucuronidation or sulfation followed by biliary excretion. Acylglucuronidation mediated by UGT2B7 and oxidation mediated by CYP2C8 may also play a role in Flamydol metabolism. CYP3A4 is responsible for the formation of minor metabolites, 5-hydroxy- and 3’-hydroxy-Flamydol. In patients with renal dysfunction, peak concentrations of metabolites 4'-hydroxy- and 5-hydroxy-Flamydol were approximately 50% and 4% of the parent compound after single oral dosing compared to 27% and 1% in normal healthy subjects.

Flamydol is eliminated through metabolism and subsequent urinary and biliary excretion of the glucuronide and the sulfate conjugates of the metabolites. Little or no free unchanged Flamydol is excreted in the urine. Approximately 65% of the dose is excreted in the urine and approximately 35% in the bile as conjugates of unchanged Flamydol plus metabolites. Because renal elimination is not a significant pathway of elimination for unchanged Flamydol, dosing adjustment in patients with mild to moderate renal dysfunction is not necessary. The terminal half-life of unchanged Flamydol is approximately 2 hours.

The ER doctor prescribed this medicine to me for minor knee pain. After taking it for 5 days I started getting an upset stomach and headaches and extreme fatigue. After 7 days I had extreme bloating, cramping and bloody diarrhea. I cant stay out of the bathroom. I stopped taking it yesterday morning and I’m even worse today. Can you take Flamydol and Ibuprofen together?

No, these drugs should not be used together, as they are from the same class, thus their effects will become additive increasing the risk of side effects such as bleeding and gastrointestinal ulcers to happen.

If you need additional medicine for your pain relieving treatment, and you already use Flamydol or ibuprofen consult your doctor. Your doctor may prescribe you acetaminophen of 500 mg as additional treatment, because it is much safer to be combined with ibuprofen or Flamydol.

Serious allergic reaction
