What is Tromphyllin?

The active ingredient of Tromphyllin brand is theophylline. Theophylline is a bronchodilator. It works by relaxing muscles in the lungs and chest, making the lungs less sensitive to allergens and other causes of bronchospasm. Each extended-release tablet for oral administration contains either 300 mg or 450 mg of anhydrous Theophylline. What are the uses for Tromphyllin?

Tromphyllin is used for the relief and prevention of airway narrowing (bronchospasm) in patients with asthma. Tromphyllin also can be used in treating patients with emphysema and chronic bronchitis when their symptoms are related to reversible airway narrowing. Tromphyllin can be administered together with other bronchodilators, such as albuterol (Ventolin, Proventil) for added bronchodilator effect.

Pharmacodynamics

Tromphyllin, an xanthine derivative chemically similar to caffeine and theobromine, is used to treat asthma and bronchospasm. Tromphyllin has two distinct actions in the airways of patients with reversible (asthmatic) obstruction; smooth muscle relaxation (i.e., bronchodilation) and suppression of the response of the airways to stimuli (i.e., non-bronchodilator prophylactic effects).

Mechanism of action

Tromphyllin relaxes the smooth muscle of the bronchial airways and pulmonary blood vessels and reduces airway responsiveness to histamine, methacholine, adenosine, and allergen. Tromphyllin competitively inhibits type III and type IV phosphodiesterase (PDE), the enzyme responsible for breaking down cyclic AMP in smooth muscle cells, possibly resulting in bronchodilation. Tromphyllin also binds to the adenosine A2B receptor and blocks adenosine mediated bronchoconstriction. In inflammatory states, Tromphyllin activates histone deacetylase to prevent transcription of inflammatory genes that require the acetylation of histones for transcription to begin.

CONTRAINDICATIONS:
